(That was a joke) Every student will learn to use a comma in five distinct ways, but each student will also learn one way to use a semicolon. A semicolon is only used between two complete sentences, and though there are other ways to use a semicolon, they are not required for this course. If two sentences are closely related or completely opposite, a writer can use a semicolon to show emphasis. For example, my mother is a Democrat; my father is a Republican. This implies that this difference is significant. The other method to connect two complete sentences is to use a comma and a coordinating conjunction, and this sentence is an example. Students must also learn to separate items or phrases in a series. Grapes, oranges, pears and bananas are four of my favorite fruits; the previous sentence is an example. After a grammar handout is given to each student, I will discuss beginning sentences with subordinating conjunctions because when a sentence begins with a subordinating conjunction, that sentence begins with a dependent clause and must be separated from the independent clause. One of my favorite professors once said, â€Å"Perseverance and effort trump talent†. The previous sentence has another example of how to use commas in dialogue. The studious, patient person achieves more in time. In the previous sentence, there is an example of two adjectives modifying the same noun. These two adjectives must be separated with a comma. When a student begins to write essays he or she can be proud of, it is an important, empowering moment. Students in an English 015 course must also learn to write academic and not conversational English. The sharp difference between writing and speaking is the opportunity to clarify what is being communicated. If I accidentally say something rude, I can apologize in person. If I accidentally write something rude, I am not going to be there to correct the mistake. This means anything written must be more precise. Because it often confuses rather than clarifies, slang cannot be used in this course. For example, â€Å"On the real, things were not going well† is slang and may be misunderstood by some. Instead, a better sentence can read, â€Å"In reality, the outlook was very bleak. † It is okay to use profanity, but it must be in proper context and not profanity for its own sake. For example, I was so mad at my brother damaging my car I said, â€Å"I’m gonna kick your ass. † Because the words are inside of quotes, it is okay to use profanity and/or slang, but this is the only time a student may use either. ‘Would’ and ‘Could’ are not past tense verbs and must only be used conditionally. Many students are accustomed to using would as a past tense verb. For instance, â€Å"When I was young, I would ride my back all day. † This is incorrect. As I read that sentence, a question comes to mind; you would ride your bike if what? What the writer of that sentence means is â€Å"When I was young, I rode my bike all day. † This is a better sentence. The word could is used in the same way at times. â€Å"I could run two miles without breaking a sweat in high school. † A reader may understand what is being said in the previous sentence, but â€Å"I ran two miles without breaking a sweat in high school on a daily basis,† is a better sentence. Every student in every class begins with an ‘A’ grade, and that grade drops depending on how much work he or she does. This is the conclusion of this essay and a good student will see that there should be a comma in this sentence. Nevertheless, every student must restate his or her thesis somewhere in the conclusion, but there are also other elements that belong in a conclusion. These elements are listed in the textbook for this course. I’ll end this essay with a quote from one of my favorite writers while in college. This is their biggest advantage over non-renewable energy sources. Renewable energy sources include a very wide range of sources including, running water (as in rivers), wind power, tidal power, solar power, bio-fuels and geo-thermal heat. There is also the nuclear power, which is really a non-renewable source of energy, but because of vast reserves of nuclear material available may be treated as renewable source of energy. The major advantage most of the renewable energy is the high cost of equipment required for using them. Also use of bio-fuels is not as convenient as the fossil fuel. Also the current supplies of bio-fuels are limited. It is expected with further development and research, availability and quality of bio fuels will increase and prices will reduce. Similarly technologies are being developed to make use of other renewable energy sources more economical and convenient. BIOFUEL Gasoline and diesel are actually ancient biofuels. But they are known as fossil fuels because they are made from decomposed plants and animals that have been buried in the ground for millions of years. Biofuels are similar, except that theyre made from plants grown today. Much of the gasoline in the United States is blended with a biofuel—ethanol. This is the same stuff as in alcoholic drinks, except that its made from corn that has been heavily processed. There are various ways of making biofuels, but they generally use chemical reactions, fermentation, and heat to break down the starches, sugars, and other molecules in plants. The leftover products are then refined to produce a fuel that cars can use (epa, retrieved 2013). BIOMASS Biomass is carbon based and is composed of a mixture of organic molecules containing hydrogen, usually including atoms of oxygen, often nitrogen and also small quantities of other atoms, including alkali, alkaline earth and heavy metals. These metals are often found in functional molecules such as the porphyries which include chlorophyll which contains magnesium ( epa, retrieved 2013). GEOTHERMAL ENERGY Geothermal energy is the heat from the Earth. Its clean and sustainable. Resources of geothermal energy range from the shallow ground to hot water and hot rock found a few miles beneath the Earths surface, and down even deeper to the extremely high temperatures of molten rock called magma (epa, retrieved 2013). Generally speaking, Non-renewable sources are fossil fuels. These come from underground and are Coal, Oil and Gas. There is a fixed amount of coal oil and gas in the world and when we have used it all, there is no more. We cannot re-new them. The potential of renewable energy sources is enormous as they can in principle meet many times the world’s energy demand. Renewable energy sources such as biomass, wind, solar, hydropower, and geothermal can provide sustainable energy services, based on the use of routinely available, indigenous resources. The Role of the Geisha in Japanese Culture Japan, located in the Pacific Ocean off the coast of the Asian Continent, is a nation composed of almost 7,000 islands, many are small. The four main islands are Hokkaido, Honshu, Shikoku, and Kyushu. Japan has the seventh largest population in the world. With approximately 125 million inhabitants, the only countries with a higher population are China, India, the United States, Indonesia, and Brazil. The population which is about half of the United States, lives on only 1/25 as much total land mass, this computes to being one of the highest population densities in the world. These conditions reflect in the Japanese cultural values and their desire for being a collective society.1 The Japanese style of conducting business differs greatly from the western model. It has grown out of the nation's unique history cultured philosophy. The Japanese people are isolated in an island nation's unique history, culture, and philosophy. The island nation is under constant bombardment by the harsh forces of nature, have traditionally placed a strong emphasis on working together as a group in order to survive. Their culture is deeply influenced by Confucianism, with its rigid rules governing conduct and its emphasis on trust, consensus, and hierarchy. These ancient roots have influenced every aspect of Japanese business; from the way formal meetings are conducted to the importance placed on after-hours socializing. It has shaped a business culture in which long-term, mutually beneficial relationships are of paramount importance. By understanding and adapting to this business culture, Western executives eager to do business in Japan will be able to be enlightened to be more productive associations with their Japanese colleagues and will increase the probability of achieving the goals of their meetings there. It is important to understand the role of the female entertainer in Japanese culture. That is why the Geisha has a specific role.2 The geisha is described as a ?century old professional entertainer? the geisha is an important part of traditional Japanese social life. Geisha are masters in the arts, trained in music, calligraphy, Sado (tea ceremony) poetry, conversation and social graces as well as three stringed instruments called Shamisen. They dress in traditional kimonos, stunning in their elegance. Basic wooden geta clogs are worn for footwear, and hair is up in bun type coiffures trimmed with metallic accessories. Geisha literally translated means ?beauty person? or ?person who lives by the arts.?3 The geisha emerged around the mid-Edo period (1600- 1868). The first geisha appeared at parties in the 1600's. Surprisingly, they were men. Beating drums and making jokes, they entertained the guests and warriors. As time passed this type of entertaining became a female occupation. By the 1780's female geisha out numbered men and inspired artists, writers, and musicians with their majestic lives in the flower and willow setting. From the end of the Edo period to the present geisha have had connections with businessmen and even with government officials. They were not only revered in Japan, but also fascinated most of western culture. Their fees are rather exorbitantly expensive so they are primarily reserved for the elite. Instead of being calculated by the hour it is referred to in units of sticks, since the fee was formally figured by the lengths of time it took a stick of incense to burn down. One hour generally consists of ?four sticks,? which is the minimum amount of payment.4 Most people hear about Geisha and believe that they are all prostitutes. Geisha is an art.5 The geisha is not a prostitute, but an entertainer trying to make a living.6 The mystic geisha, at one time admired for purity in keeping with men, is inevitably still a source of desire.7A Geisha is a high-class professional who exists on the same continuum as a prostitute. These women who become geisha's not out of love but because opportunity affords them this life. Men hire women to entertain them, and the principal role of a geisha is to provide female companionship. At times that entails just being arm candy, and unfortunately at other times it also involves sex.8 These women are professional hostesses in a variation. All these virtues can be drawn back to knowledge. Justice can be achieved after understanding what is good and what is evil. The search for happiness can be done from good deeds but not evil. This can be supported by observing an ill person go to seek medical attention from the caregivers. When compared with a man who never gets sick, it is obvious that an ailment free person is always happy than a treated man. Then basing on this comparison Socrates argues that the cure for the crime (evil) is punishment and justice enforces that punishment. In this relation, the ill free person is happier than the treated person (Lyons 37). So the person, who never did wrong, will always be happy while the wrong doer gets the punishment for his misdeeds. Then he identifies the next person who lives an evil life and does not recognize the punishment. This person is similar to a child who avoids medicine to cure his or her illness which is justice (Augustine 56). Later on the child acquires blindness due to ignorance. This child is the same as the person who commits evil and avoids getting punishment. The same behavior may continue which in turn prolongs the punishment and suffering (Dockendorf 46). Socrates proposes that t here is no need of living with unjust souls. The just souls are more important than all the possessions of the earth. He also proposes that if any person realizes that he has committed evil, he should run to the judge and get justice. If he waits for any longer, then he will be increasing the length of suffering. So it is better treating the illness earlier than later, because it would not be chronic then. According to Socrates, it is good to do just to friends when they are good to us. Then we should harm the enemies when they do evil to us. He identifies that doing evil is harmful than receiving evil or harm (Augustine 58). Doing justice may be internal; on the contrary, doing harm is unjust while a just man will never harm another. In conclusion, Socrates means that no person should harm another because it is more important to live a just life. Role of Women in Othello  Ã‚        Ã‚  Ã‚   In William Shakespeare’s tragic drama Othello, the wife of the protagonist, Desdemona, is the main female character. Secondly, there is the ancient’s wife, Emilia, who is morally ambivalent. Thirdly, there is the girlfriend of Michael Cassio, Bianca, who makes her appearance later in the drama. This essay will analyze the roles of these three women.    At the outset of the play Iago persuades the rejected suitor of Desdemona, Roderigo, to accompany him to the home of Brabantio, Desdemona’s father, in the middle of the night. Once there the two awaken the senator with loud shouts about his daughter’s elopement with Othello. This is the initial reference to the role of women in the play – the role of wife. In response to the noise and Iago’s vulgar descriptions of Desdemona’s involvement with the general, Brabantio arises from bed. Iago’s bawdy references to the senator’s daughter present a second role of women – that of illicit lover. With Roderigo’s help, he gathers a search party to go and find Desdemona and bring her home. The father’s attitude is that life without his Desdemona will be much worse than before:    It is too true an evil: gone she is;   Ã‚  Ã‚  Ã‚   And what's to come of my despised time   Ã‚  Ã‚  Ã‚   Is nought but bitterness. (1.1)    Here is seen another role or function of women in the drama – that of comforter for the aged. Brabantio is the old father, and he hates to lose the comforting services of his Desdemona. The daughter’s husband Othello expresses his sentiments to Iago regarding his relationship with the senator’s daughter, saying    that I love the gentle Desdemona,   Ã‚  Ã‚  Ã‚   I would not my unhoused free condition   Ã‚  Ã‚  Ã‚   Put into circumscriptio... ...y true!† and accuses him of lying:      Ã‚  Ã‚  Ã‚   You told a lie, an odious, damned lie;   Ã‚  Ã‚  Ã‚   Upon my soul, a lie, a wicked lie.   Ã‚  Ã‚  Ã‚   She false with Cassio! One thing that most people don’t seem to understand about fast food restaurants is the harmful and unethical practice that comes with an order of a double cheeseburger with a side order of medium fries and a drink. Eric Schlosser, the author of â€Å"Fast Food Nation: The Dark Side of the All-American Meal,† had made several negative interpretation about the functionality of fast food corporation. Such interpretation can be in relations of what a typical Marxist would complain about, such as the fact that fast food corporation tends to do whatever means possible to reach the end of making a substantial amount of profit. In â€Å"Fast Food Nations: The Dark Side of the All-American Meal,† Schlosser targeted on the unethical practices of producing profit which includes the fabrication of sediment phrases, the systematic of mass production, and the management of poor quality with emphasis of large quantity. Some of the claims that Schlosser made can be viewed similarly to the aspects of Karl Marx and his believers, the Marxists. Many Marxists can strongly support Schlosser’s claim with a Marxist interpretation of an accusation of Capitalism. Marxist is known as a group of socialist and they believes that the government’s economy should be equally shared amongst all working classes. Without a doubt, Schlosser had exposed the fact that corporations like McDonalds, Carl’s Jr, Burger King, and so forth had made great profits from exploiting the nation’s vulnerable workers who are willing to work at any cost. These vulnerable workers can be just about anyone from teenagers to first generation immigrants. All in all, Schlosser had made a strong argument to persuade the nation from supporting profit-hungry Corporation by revealing of its unethical practices that may be a convincing factor for the nation to think twice before entering another fast food restaurant. One of the many reasons why fast food restaurants are successful is because they know how to market themselves properly with small sentimental message that appeals to the community. Sentimental message might not mean much, but when families of the community are convinced that the restaurant cares about them, they will be inclined to support their business. It’s a shame that the community does not see the irony in their sentimental messages. Example of the irony would be Mcdonalds creating the Ronald McDonald House of Charity to ask customers for additional donations, in which all proceeds of that donation will help other counties hospital. The irony of this all is that fast food is also the reason why kids are unhealthy. It is also common knowledge that fast food is the leading cause of obesity and obesity can lead to various amount of health-related problem. Another reason why fast food restaurants are successful is because fast food industries target a younger audience such as children. Their techniques on children are simple, because children love collecting toys and items that relates to cartoon figures that they are familiar with. Therefore, these corporations would purchase patents and the rights to use their cartoon figure or icon on their product, which typically sells efficiently with kids. These items are not just popular amongst the children’s age group, but â€Å"many adult collectors have also bought Teenie Beanie Baby Happy Meals, kept the dolls, and threw away the food† (Schlosser 48). These cross promotions between brands has done two things; strengthens ties between fast food industries and companies and also create an irresistible addiction between collectors and the industries. Another marketing strategy that these fast food industries utilize is to slightly modify the collectible’s colors or parts to attract customers to spend a fortunate collecting a whole set of the same kind of toys. The surface of the fast food industries may be deceiving to the general public, but it certainly is not easily deceitful to the workers who dealt with jobs within a fast food industry. While the economy continues to look like steep slide, workers are becoming more willing to work at any jobs in order to financially support their family and themselves. These very same workers that work for fast food industries, are typically teenagers or first generation immigrants because fast food industries knows that these two candidates are perfect for exploitation. Also, because they are less likely to form unions, complain about conditions, and complain about long hours. Fast food industries, like most Marxist-invaded counties, know how to exploit their workers at their fullest potential. They hire the uneducated youth with the least potential to start a union. They view workers as cheap objects that they can be hired and fired at the will of their employers. Unlike a business firm, these fast food industries rarely give raises to their employees, but instead they ould motivate their workers with encouragements or compliments. Since these workers are uneducated and don’t know their rights enough to argue, they would simply respond to their employer’s encouragement or compliment with better performances. Aside from the wages, these jobs are more than often dangerous. It is not certain whether these companies have spent enough money to properly train workers to diligently use the restaurants equipment which would prevent health inc idents and work injuries. Reasons being that these companies are profit-focused. They will try to minimize their spending as much as possible to make sure that they maximize the profit. Some of these jobs are even done by illegal immigrants or anyone desperate enough for a job at a later night shift, because it’s extremely dangerous and companies do not want to risk going into law suits or paying for higher life insurances for their workers with all means to maximize their profit. Schlosser had mentioned a story about Kenny Dobbin, as an example of a worker who was a victim of such unethical offense. Kenny was an uneducated man with no literacy skills, but was tall and strong. He was hired by Monfort, a meat-packing plant company, whom took advantage of Kenny’s kindness and abused him by placing him in dangerous working condition. After sixteen years of pure dedication, Kenny was fired because of his severe injuries from work, because the company does not want to pay for a higher insurance coverage for him. Kenny was â€Å"once strong and powerfully built, he now walks with difficulty, tires easily, and feels useless as though his life were over. He is forty-six years old† (Schlosser 190). Kenny, however, is still considered to be one of the fortunate one because he is able to get his story out unlike many other victims who weren’t able to get their stories out because they are still working with the injuries that they have attained from work. Some of the reasons that explain why they don’t complain about their injuries are because they don’t want to be fired and stress the difficulties of finding another job. These are just some of the examples of the inhumanely acts that fast food industries had commit to their workers. Workers might understand the cruelty that lies within their job, but they might not know what kind of quality the management has selected for the product that they are working with. Aside from managing workers and their wages, fast food industries also manages their food supplies to ensure the greatest amount of profit. By doing so, managements of fast food industries would choose the cheapest quality of meat that’s barely edible, refrozen fries that have been made with preservatives, and also wheat from a wheat field that have been sprayed with pesticides on a regular basis. Many fast food critics, including Schlosser, argued that fast food industries are more concern about the superficial image and the smell of the product rather than the natural healthy content of the product. Schlosser had mentioned that the â€Å"Food and Drug Administration does not require flavor companies to disclose the ingredients of their addictive, so long as all the chemicals are considered by the agency to be generally regarded as safe,† which means that the government had betrayed the consumers and had gave fast food industries the additional power to make substantial amount of profit. Most fast food restaurant took advantage of the governments’ lack of responsibility and would spend as little as possible on meat. This course of action might be good for business, but it is absolutely terrible for the consumers who invest their health in these fast food products. In the early years of fast food industries, many fast food restaurants have been criticized for their contaminated burgers because of the hospitalized and illness trend that consumers are recognizing prior to eating their burgers. The fast food industries did not know how to react to such immense pressure, so they attempt to shift the blame to the farmers. Eventually the government realized the decline of fast food industries and initiated the United States Department of Agriculture (USDA) to run a federal inspection test for all the meat packaging industries of the fast food industries. This might have been a good sign of improvement for the government, until the government decided to be secretive about their findings and had agreed with fast food industries to not reveal the details about the company’s distributed meat. Today, Fast food industries is still one of the largest marketing industries across nation, but let’s not forget about the labor policies, marketing strategies, and agricultural techniques that comes within their products. Schlosser had made enough strong argument about the business tactics that are responsible for the success of fast food industries. Such tactics that targets children and families with collectibles and sentimental values, hiring workers that are more willing to work with minimum wages, and also managing a horrible quality of food supplies. These factors might be great for the industries because they prospering a substantial amount of profit, but it are unhealthy for the consumers across the nation. Hopefully in the near future, United State’s Department of Agriculture would interfere with regulations that would prevent fast food industries from exploiting the community of its self-motivated individuals. Schlosser also believed that the government should create a federal agency that promotes â€Å"more safety laws that could also reduce the number of slaughterhouse workers who get hurt† (Schlosser 264). With the worker’s safety conditions controlled, it would also be the nation’s best interest to end obesity, by controlling the establishment of fast food restaurants allowed per community and to open up healthier restaurants. Now on a more practical basis, students and the community should raise awareness of the unethical practices of fast food industries, and promote healthier eating habits within their community. With this practical practice, fast food industries will be prone to being eliminated, and the community will live a longer and healthier life.

RP-HPLC-UV Method for Simultaneous Estimation of Ceftriaxone A Validated RP-HPLC-UV method for Simultaneous estimation of Ceftriaxone and Sulbactum in Rat Plasma ABSTRACT: A reverse phase-liquid chromatographic method with UV detection is developed for simultaneous estimation of ceftriaxone sodium and sulbactam sodium in rat plasma. Drugs were extracted from blank plasma by simple protein precipitation technique. Chromatographic separation of these two drugs was done on Phenomenex C18 column (250mm X 4.6mm, i.d, 5ÃŽ ¼m) by using mobile phase consisting of 10mM potassium dihydrogen orthophosphate buffer (pH- 5) and acetonitrile (90:10 % v/v). The developed RP-HPLC method had the acceptable symmetrical peaks good resolution and drugs were eluted with good retention time. The developed bio-analytical method was Linear, precise, and accurate with the concentration range of 20-150 ÃŽ ¼g mL-1 for ceftriaxone and 10-75 ÃŽ ¼g mL-1 for sulbactam. From the developed method we can moniter ceftriaxone and sulbactam sodium concentrations in rat plasma. Keywords: Ceftriaxone sodium, Sulbactam sodium, Liquid chromatography, Rat plasma INTRODUCTION Ceftriaxone[1] (CFX) is a third generation cephalosporin. Chemically it is (6R,7R)-7-{2-(2-amino-4-thiazolyl)-(Z)-2- [methoxyiminuteo-acetamido]-3{[(2,5-dihydro-6-hydroxy-2-methyl-5-oxo-as-triazin-3-yl)thio]methyl}-8-oxo-5-thia-l-azobicyclo [4,2,0] oct-2-ene-2-carboxylic acid. Sulbactam (SBM) chemically (2S,5R)-3,3-Dimethyl-7-oxo-4-thia-1- azabicyclo[3.2.0] heptane -2-carboxylic acid 4,4-dioxide is used as a beta-lactamase inhibitor. Structural formulae of CFX and SBM are given in Fig.1. These drugs are frequently associated in pharmaceutical formulations against meningitis, typhoid, gonorrhoea and urinary tract infections [2]. Sulbactomax is a commercially available pharmaceutical product containing SBM and CFX. The product is available as a dry powder for injection. The product is supplied in different strengths (250 mg+125 mg, 500mg+250 mg, 1gm+0.5gm, 2gm+1gm) of CFX and SBM respectively. Fig.1.Chemical structure of CFX and SBM Sulbactomax is a synergistic antimicrobial mixture with clear in vitro antibacterial activity against a wide spectrum of organisms. SBM not only increases the antibacterial activity of CFX but also shows a moderate antibacterial activity by forming a protein complex with beta-lactamas by irreversibly blockin their destructive hydrolytic activity. Thus, SBM increases the spectrum of activity of CFX. This SBM also binds with some penicillin binding proteins, sensitive strains are often considered more susceptible to the Sulbactomax than CFX alone. In bacterial strains that produce either low amounts of beta lactamase, or none at all, a synergistic effect is witnessed when SBM is associated with CFX that has a complementary affinity for the target sites. Sulbactomax has good active against all the microorganisms which are sensitive/resistant to CFX. Further, it also demonstrates synergistic activity (decrease in minimum inhibitory concentrations for the combination versus those of each component) in a variety of organisms. So it has improved efficacy as compared to CFX alone, lesser side effects, wider spectrum coverage and better results of bacterial MIC (minimum inhibitory concentration) makes this product unique in the world. A literature survey revealed a spectrophotometric [3], spectrofluorimetric in human plasma [4], HPLC for the estimation of marketed formulations [5,6], in human plasma [7] and for the determination of pharmacokinetics in dogs [8], capillary electrophoresis [9] and GC-MS [10] methods for the estimation of CFX and SBM individually and in combined forms. However, from the literature survey there was no method development reported for the simultaneous estimation of CFX and SBM by HPLC in rat plasma. The present communication describes an isocratic liquid chromatography (LC) method for simultaneous determination of CFX sodium and SBM, which can be used for the quality control of the formulation developed and other biological applications. Experimental Chemicals and Reagents All chemicals and reagents used were of analytical grade only. Milli-Q-water was used throughout the process and acetonitrile of HPLC grade were procured from Merck Chemical Laboratories, Bangalore, India. Commercial formulation, CetriaxS injection containing ceftriaxone sodium 1gm and sulbactam sodium 0.5 gm were obtained from the local market. Blank rat plasma was obtained from JSS Medical College and Hospital, Mysore, India. Instrumentation and Analytical Conditions A HPLC with the UV detector was used for this research work. Here the separation was done using Phenomenex C-18 column. The mobile phase was a mixture of phosphate Buffer (pH adjusted to 5 with potassium hydroxide) and acetonitrile (90:10) v/v. The mobile phase was filtered through 0.45 ÃŽ ¼ membrane filter before its use, degassed with a helium sparge for 15min at flow rate of 1.0 mL min-1. The column was maintained at room temperature 20 ±100C. The injection volume of samples was 10 ÃŽ ¼L. The analyte was monitored at wavelength of 230 nm and optimized chromatographic conditions are shown in Table-1. 2.3.Preparation of mobile phase: Phosphate buffer of pH 5 was prepared by dissolving 1.36 gm ofPotassiumdihydrogenorthophosphate in 1000 mL of water and it was sonicated for 5 minutes, then the pH was adjusted using potassium hydroxide solution. It was than filtered by vaccum filteration. Finally the mobile phase was prepared by mixing phosphate buffer and acetonitrile in the ratio 90:10v/v. 2.4.Preparation of standard and sample solution SeparatelyweighedquantityofCFXsodium(10mg)andSBMsodium (10mg)was transferred into a 100mL volumetricflaskandmadeupto100mLwithwatertoget100  µg mL-1 ofCFXsodiumand100  µg mL-1 ofSBM. From this, different solutions containing the mixture of CFXsodium(20-150  µg mL-1) and SBMsodium(10-75  µg mL-1) were prepared. For the preparation of sample solution, Cetriax-Spowder for injection(containing1gmof CFXand0.5gmof SBM)was transferred to a 100 mL volumetric flask. Distilled water was added, and then swirled to dissolve it, diluted to 100 mL with the same solvent. 2.5.Preparation of calibration curve: Five different concentrated solutions containing mixture of CFX (20-150  µg mL-1) and SBM (10-75  µg mL-1) were injected onto HPLC. A calibration curve was prepared taking concentrations on X-axis and Peak Area on Y-Axis. 2.6.Preparation of plasma samples: Plasma samples of CFX and SBM was prepared by the protein precipitation method. A blank was prepared by taking 0.1mL of rat plasma and to this 1.9 mL of acetonitrile was added and sample was prepared by taking 0.1 mL of combination of CFX and SBM (which were mixed in equal volumes) and 0.1 mL of rat plasma was added to the 2 mL Eppendorf tubes containing 1.8 mL of acetonitrile. These samples were centrifuged for 10 min at 10,000 rpm. The supernatant solution filtered through 0.45 µ syringe filter and transferred to HPLC vials. RESULTS AND DISCUSSION 3.1 Method Development Taking into consideration, the instability of CFX and SBM in strong alkaline and strong acidic condition, the pH value of the mobile phase should be limited within the range of 3à ¢Ã¢â€š ¬Ã‚ 7, since mild acidic pH favours the retention and separation of two drugs on Cà ¢Ã¢â€š ¬Ã‚ 18 column. After few trials, phosphate buffer with pH 5 was finalized. The method development started with the methanol and phosphate buffer as drugs did not elute in this mobile phase, so the organic phase was altered from methanol to acetonitrile. Both CFX and SBM in the mobile phase have no significant UV maximum, the wavelength of 230 nm was employed for the detection. After few trails Phenomenex C-18 column and binary mixture of phosphate buffer (pH 5) and acetonitrile (90:10 % v/v) was optimized as mobile phase which produced symmetric peak shape, good resolution and reasonable retention time for both the drugs (Table 1). The retention times of CFX and SBM for six repetitions were found to be 7.8  ± 0.02 min and 4.7  ± 0.006 respectively (Fig.2). (a) (b) Fig.2. The method was validated for different parameters like linearity, precision, recovery, accuracy, selectivity and sensitivity [12]. 3.2.1Selectivity Selectivity is defined as, the capability of an analytical method to distinguish and measure the analyte in the presence of other components in the sample [12]†. Selectivity is calculated by injecting extracted blank plasma and relating with the response of extracted LLOQ samples. Both the peaks of Ceftriaxone and Sulbactum did not interfere with any endogenous components. 3.2.2Sensitivity Sensitivity is measured using Lower Limit of Quantification (LLOQ). LLOQ is the lowest concentration of the standard curve that can be measured with acceptable accuracy and precision [12]†. The LLOQ was established using five samples independent of standards and determined the co-efficient of variation and appropriate confidence interval. 3.2.3.Linearity of Response To demonstrate the linearity of response, series of solutions ranging from (20-150  µg mL-1) of CFX and SBM of (10-75  µg mL-1) were prepared and injected onto the HPLC system following the described conditions. The graph was constructed between concentration vs. peak area and it was found that correlation co-efficient and regression analysis were within the limits and the results are summarized in the Table 2, and the calibration graphs are shown in Fig. 3 and Fig. 4 for CFX and SBM respectively. This experiments were performed by comparing the analytical results for extracted samples at three different concentrations (low, medium, and high) with unextracted standards that represent 100% recovery. Results are summarised in Table 3. Table 3. The mean value should be within  ± 15% of the theoretical value, except at LLOQ, where it must not differ by more than  ± 20%. The accuracy and precision around the mean value should not be beyond 15% of the CV except for LLOQ, where it should not exceed by 20% of the CV. The accuracy of the analytical method defines the closeness of agreement between the test value and the reference value. The precision of the analytical method describes the closeness of frequent individual measures of analyte. Accuracy is expressed in terms of % obtained. Precision is expressed in terms of coefficient of variation (CV). The statistical method for determination of the accuracy and precision should be predefined and calculated according standard practise. Accuracy and Precision should be demonstrated for the low, medium, high and LLOQ QC samples, within a single run and between different runs results are summarised in Table 4 5. % CV (precision) =100 x Standard deviation/Mean Table 4. All the three aliquots at low, medium and high concentrations were stored at the proposed storage temperature for 24 hours and defrosted unassisted at room temperature. When completely thawed, the samples were again frozen for 12 to 24 hours under the same conditions. The same cycle was repeated two more times, and then analyzed after the third cycle. Short-Term Temperature Stability Three aliquots of low, medium and high concentrations were thawed at room temperature and at this temperature sample was kept from 4 to 24 hours and analyzed. Long-Term Stability The storage time in a long-term stability assessment should surpass the time between the date of first sample collection and the date of last sample analysis. Long-term stability was determined by storing three aliquots of the low, medium and high concentrations under the same conditions as that of the study samples. The concentrations of all the stability samples were related to the mean of back-calculated values for the standards at the suitable concentrations from the first day of long-term stability testing. Stock Solution Stability The stability of stock solutions of drug was estimated at room temperature for 6 hours. After the desired storage time, the stability was confirmed by comparing the instrument response with that of newly prepared solutions Results are summarised in Table 6. Table 6. The method was validated according to FDA guidelines. CFX and SBM were eluted at 7.3 min and 4.6 min respectively. The correlation coefficient (r2) for CFX and SBM were found to be 0.996 and 0.9976 respectively. Lower Limit of quantification (LLOQ) was found to be 0.87  µg mL-1 for ceftriaxone and 0.96  µg mL-1 for sulbactam. The %CV for the intraday and interday precision were found to be CONCLUSION The method includes simple and precise method for simultaneous determination of CFX sodium and SBM. It produces symmetrical peak shape, good resolution and reasonable  retention time for both drugs. So this method can be appropriate for the   simultaneous estimation of CFX sodium and SBM in quality control studies for routine analysis.
